Quantitative MRI assessment of matrix development in cell-seeded natural urinary bladder smooth muscle tissue-engineered constructs

چکیده
INTRODUCTION Cell-seeded natural scaffolds derived from acellular matrices (ACMs) hold great promise for tissue-engineering large soft-tissue organs such as the urinary bladder and heart. The ACM has advantages of biological recognition and structural and biochemical similarities with the target tissue that are not easily reproduced using conventional synthetic or tissue-mimicking materials [1]. However, experience with cell-seeded natural scaffolds is relatively young, and little is known about cell-matrix interaction or its influence on MRI characterization. This study investigates a battery of quantitative MRI methods to monitor cell-matrix interaction and matrix development in a model of smooth muscle cell-seeded urinary bladder ACM.

Does the Mesenchymal Stem Cell Source Influence Smooth Muscle Regeneration in Tissue-Engineered Urinary Bladders?
A variety of tissue engineering techniques utilizing different cells and biomaterials are currently being explored to construct urinary bladder walls de novo, but so far no approach is clearly superior. The aim of this study was to determine whether mesenchymal stem cells (MSCs) isolated from different sources, (bone marrow [BM-MSCs] and adipose tissue [ADSCs]), differ in their potential to reg...

Objective: Vascular-derived cells represent an established cell source for tissue engineering of cardiovascular constructs. Previously, cell isolation was performed by harvesting of vascular structures prior to scaffold seeding. Marrow stromal cells (MSC) demonstrate the ability to differentiate into multiple mesenchymal cell lineages and would offer an alternative cell source for tissue engine...
متن کاملEnhancing mechanical properties of tissue-engineered constructs via lysyl oxidase crosslinking activity.
A number of strategies have been investigated to enhance the mechanical stability of engineered tissues. In this study, we utilized lysyl oxidase (LO) to enzymatically crosslink extracellular matrix (ECM) proteins, particularly collagen and elastin, to enhance the mechanical integrity of the ECM and thereby impart mechanical strength to the engineered tissue. Vascular smooth muscle cells (VSMCs...
متن کاملTerminal urothelium differentiation of engineered neoureter after in vivo maturation in the "omental bioreactor".
OBJECTIVE Long ureteric defects may theoretically be repaired with the use of tissue-engineered neoureter. However, attempts to construct such a neoureter in animal models have failed because of major inflammatory response. Avoidance of such inflammation requires a well-differentiated urothelium. We investigated whether omental maturation of a seeded construct in a pig model could achieve termi...
متن کاملTissue-engineered autologous bladders for patients needing cystoplasty.
BACKGROUND Patients with end-stage bladder disease can be treated with cystoplasty using gastrointestinal segments. The presence of such segments in the urinary tract has been associated with many complications. We explored an alternative approach using autologous engineered bladder tissues for reconstruction. METHODS Seven patients with myelomeningocele, aged 4-19 years, with high-pressure o...
